The Effect of Sambucus williamsii Hance(SWH) on Osteoporosis Induced by Ovariotomy in Rats


Abstract
Osteoporosis is a disease characterized by bone loss and morbidity with osteoporotic fracture is increasing in elderly.
This study was performed to evaluate the effect of Sambucus williamsii Hance on the bone mass and its related factors, using ovariotomized rat as an estrogen-deficient animal model. The model rats of osteoporosis showed a significant decrease in bone density, bone ash density, calcium content of femur bone.
At the 7th day after operating ovariotomy, rats were administered with Sambucus williamsii Hance per orally, and continued for 10 weeks. And osteoporosis related parameters were determined to investigate the effect of Sambucus williamsii Hance.
The results were summerized as follows:
1. The level of serum T_3, T_4 in control group showed no significant change in comparison with sham, but that Sambucus williamsii Hance-treated showed significant decrease in comparison with control.
2. The level of serum PTH in control group showed significant increase in comparison with sham, but that Sambucus williamsii Hance-treated showed significant decrease in comparison with control.
3. The level of serum phosphorus in control group showed no significant change in comparison with sham, but that Sambucus williamsii Hance-treated showed significant increase in comparison with control.
4. The level of ionized calcium in control group was slightly decreased in comparison with sham, which showed no efficacy. but that Sambucus williamsii Hance-treated showed no significant change in comparison with control.
5. The level of total protein in control group showed significant decrease in comparison with sham, but that Sambucus williamsii Hance-treated showed significant increase in comparison with control.
6. The level of total cholesterol in control group showed significant decrease in comparison with sham, but that Sambucus williamsii Hance-treated showed significant increase in comparison with control.
7. The level of serum ALP in control group showed significant increase in comparison with sham, but that Sambucus williamsii Hance-treated showed no significant change in comparison with control.
8. The level of serum ALT, AST in control group was slightly increase in comparison with sham, but that Sambucus williamsii Hance-treated showed no significant change in comparison with control.
9. Body weight in control group showed significant increase in comparison with sham, but that Sambucus williamsii Hance-treated showed significant decrease in comparison with control.
10. In hematology, the level of RBC(P<0.001), Hb(P<0.05), Ht(P<0.01) in the Sambucus williamsii Hance-treated group showed siginificant increase.
11. In the experiment of protein reactivation related to bone formation, the Sambucus williamsii Hance-treated activated BMP-2 reactivation which was inhibited by 10^-7M
dexamethasone.
According to the above these results, Sambucus williamsii Hance has shown to be capable of preventing and curing osteoporosis caused by old-aged and postmenopause.
